He married Susannah Baxter. Susannah was born in 1776; daughter of Susannah Benton and Thomas Baxter.
The first record of Susannah Hopkins is the 1799 Will of George Vinson Benton. Susannah is noted as "child of his sister Susannah Benton".
Susannah Hopkins is listed as heir to George Vinson Benton's tract of land called "Suffolk" in Queen Anne's County Maryland on April 15, 1807 land records.
She is noted as "Susannah Hopkins".
James & Susannah had the following children:

Susannah Baxter Hopkins died in 1855 at age 80.
They were buried in the Hopkins Family Graveyard on their property.
The headstones were later removed and are now in the Stevensville Cemetery.
